The limit is PI's memory, thus depends on your model. Footprint of OS+samplerbox is around 250Mb, leaving over 725Mb on a 3B+.
USB versus SD: winner depends on types used, but both are slow
Samples don't have to be large: the high quality grand piano takes 250Mb, but most of the sounds I use are 20-80Mb. You can load multiple samples in memory within single sample set and switch between them using voices, while playing which can be convenient in gigs.
